چکیده

Iphosphamide [2 (2 chloroethylamino) 3 (2 chloroethyl)tetrahydro-2//-l ,3,2-oxazaphosphorine 2-oxide] is metabolized by mouse liver microsomes in the presence of reduced triphosphopyridine nucleotide and oxygen. When purified aldehyde oxidase is added, the initial metabolites do not appear in quantity; but a single, anionic product is present. After chemical methylation, the mass spectrum of the product of the aldehyde oxidase reaction is identical to that of 2-carbomethoxyethyl Ar,A^'-bis(2-chloroethyl)phos-

Comparative metabolism of 2-[bis(2-chloroethyl)amino]tetrahydro-2-H-1,3,2-oxazaphosphorine-2-oxide (cyclophosphamide) and its enantiomers in humans.

The comparative metabolism of the enantiomers of cyclo phosphamide and of the racemate has been studied in humans. Four patients were each given, sequentially, the racemate, the (+)-enantiomer, and its (-)-antipode. The plasma levels of parent drug and the urinary output (24 hr) of unchanged drug and of two enzymatically produced metabolites, 4-ketocyclophosphamide and carboxyphosphamide, were ...

Unexpected toxicity in patients treated with iphosphamide.

A derivative of cyclophosphamide, 2,3-(TV, TV1-bis(2chloroethyl)diamido-l, 3, 2-oxazaphosphoridinoxyd (Iphosphamide), was less toxic in experimental systems than cyclophosphamide, while it had a broader antitumor spectrum. We studied this agent in a clinical trial of 37 patients with histologically proved, far-advanced malignant disease. The study was designed mainly to determine toxicity and d...

Experimental chemotherapy of human medulloblastoma cell lines and transplantable xenografts with bifunctional alkylating agents.

A series of bifunctional alkylators were tested against the genotypically and phenotypically heterogeneous continuous human medulloblastoma cell lines, TE-671, Daoy, and D283 Med in vitro and against TE-671 and Daoy growing as s.c. and intracranial xenografts in athymic mice. Drugs tested included melphalan, cyclophosphamide, iphosphamide, phenylketocyclophosphamide, thiotepa, 1,3-bis(2-chloroe...

We now extend these observa tions and report the in vitro and in vivo sensitivity of TE-671 to sevenclassicalalkylators. Ag...

Enzymatic metabolism of cyclophosphamide and nicotine and production of a toxic cyclophosphamide metabolite.

Cyclophosphamide is converted by enzymes of mouse liver into two metabolites. Production of the first (aldophosphamide), which is uncharged, requires TPNH, is inhibited by CO, and is accomplished predominantly by the microsomal fraction. With the microsomal enzyme, the Km for Cyclophosphamide is 0.5 mM; nicotine, atropine, ephedrine, apomorphine, and cocaine are potent inhibitors. Phénobarbita...
